He was married to Ann Bayley. Doctor Samuel McMaster and Ann Bayley had the following children:

child50 i. Doctor John Thomas Bayley McMaster was born on 18 Dec 1827 in Worcester County, Md.. He was a President of Railroad between 1869 and 1883 in Eastern Shore of Virginia. He died on 27 Aug 1889 in Worcester County, Md.. He was a Maryland State Senator 1864 to 1866 in Pocomoke, Md.. He served in the military Civil War in Maryland Brigade of Volunteers (surgeon).
